ABSTRACT

Seven new triterpenoid saponins, including five ursane-type saponins, ilexchinenosides R-V (1-5), and two oleanane-type saponins, ilexchinenosides W-X (6-7), with four known triterpenoid saponins (8-11) were isolated from the leaves of Ilex chinensis. Their structures were elucidated by comprehensive spectroscopic 1D and 2D NMR and HR-ESI-MS data. Their sugar moieties were determined by HPLC analysis compared with standards after hydrolysis and derivatization. Compounds 1, 2, 4, 9 and 10 exhibited potential hepatoprotective activity against N-acetyl-p-aminophenol (APAP)-induced HepG2 cell injury in vitro.

ABSTRACT

Objective: To study the triterpenoids from the leaves of Ilex chinensis. Methods: The constituents were isolated and purified by various chromatographic methods, and the structures were elucidated by spectroscopic analysis. Results: Thirteen triterpenoid saponins were isolated from 70% ethanol extracts in the leaves of I. chinensis and identified as 3β, 23-dihydroxyurs-12-en-28-oic acid 28-O-β-D-glucopyranoside (1), quadranoside IV (2), mussaendoside R (3), monepaloside F (4), ilexoside XXVII (5), ilexoside XXXVII (6), ilexoside XXXVIII (7), ilexoside XLI (8), niga-ichigoside F1 (9), kalidiumoside D (10), chikusetsusaponin Iva (11), oblonganoside M (12), and 23-hydroxy-betulinic acid-28-O-β-D-glucopyranoside (13). Conclusion: Compounds 1, 10, and 13 are isolated from the genus Ilex for the first time. Compounds 2-8, 11, and 12 are isolated from this plant for the first time.

ABSTRACT

Objective:To optimize the extraction process of Ilex chinensis Sims by Box-Behnken response surface method. Meth-ods:The HPLC determination of pedunculoside was performed and validated. With the extraction rate of pedunculoside as the index, the influencing factors including material-liquid ratio, ethanol concentration, reflowing temperature and decocting frequency were opti-mized by Box-Behnken response surface method. Results:The optimized parameters were as follows:the material/liquid ratio was 1:10, the ethanol concentration was 70%, the ultrasonic temperature was 80℃,and the decocting frequency was three times. Conclu-sion:The HPLC method and the best extraction conditions are reliable.

Methods: The constituents were isolated and purified by various chromatographic methods, and the structures were elucidated by spectroscopic analysis. Results: Fifteen triterpenoid compounds were isolated from 70% ethanol extract in the leaves of I. chinensis and identified as 2α,3β-dihydroxy-23-norolean-4(24)-12(13)-dien-28-oic acid (1), 29-hydroxyhederagenin (2), ilexosapogenin A (3), rotundic acid isopropylidene (4), 2α,3β-dihydroxy-23-norurs-4(24)-12-dien-28-oic acid (5), 23-hydroxyursolic acid (6), 3β,23-dihydroxyursa-12,18- dien-28-oic acid (7), asiatic acid (8), 28-O-β-D-glucopyranoside siaresinolic acid (9), 28-O-β-D-glucopyranoside spathodic acid (10), 28-O-β-D-glucopyranoside pomolic acid (11), rotungenoside (12), 3β,23-dihydroxyurs-12,18-dien-28-O-β-D-glucopyranosyl ester (13), 3β,23-dihydroxyurs-12,19-dien-28-O-β-D-glucopyranosyl ester (14), and 3β,23-dihydroxyurs-12,19(29)-dien-28-O-β-D- glucopyranosyl ester (15). Conclusion: Compounds 1 and 2 are isolated from the plants of Ilex L. for the first time. All compounds are isolated from this plant for the first time.
